Comparison of the use of platelets-rich plasma, alone or combined, on the healing process in surgical wounds

Abstract

In order to evaluate the use of platelets-rich serum (PRP), alone or combined with other cellular activators, such as calcium chloride and / or thrombin, on the skin healing process in rabbits with artificially induced wounds. To do this, 10 experimental units were used to which 3 surgical incision wounds of 5 cm were made, an area of the back between T4 and T8 and two in the shoulder blades, which will be treated with PRP as a healer (T1), PRP + PRP with CaCl2 (T2) and PRP with Thrombin (T3). The healing effects of the 3 treatments were evaluated on days (d) 7 post-operation action, by microscopic evaluation of the wound and on d 15 the tissue evaluation will be carried out, with Masson’s trichromy staining methodology. The results consisted of obtaining: fibrous connective tissue, presence of fibroblast, presence of elements that mediate inflammation such as neutrophils, lymphocytes and histocytes, presence of neovascularization and complete healing; all of them coded on a semi-quantitative scale where 1 is placed if the finding was mild, 2 moderately and 3 complete. The results obtained indicated the non-existence of statistical difference (P<0.05) between the 3 treatments applied, however, all of them demonstrated an effective advance in the healing process in the skin of rabbits, by demonstrating a clear indication of acceleration of the vascularization processes and the abundance and proliferation of fibroblasts from d 7, which shortens the healing time of wounds, consolidating PRP as a therapeutic alternative in tissue repair.
